Analysis of Genetic Diversity in Onychostoma macrolepis Populations

Abstract
In order to analyse the genetic diversity and genetic differentiation of Onychostoma macrolepis. A total of 50 samples were obtained from two geographical locations (Caishi River and the River of Culai Mountain which is near Mount Tai) for analysis. Sequence analysis was performed using MEGA (version 6.0) and DnaSP (version 5.0) software. The results demonstrated that 11 haplotypes were detected in 50 sequences. There were significantly more haplotypes in the River of Culai Mountain area than in the Caishi River area. The genetic distance was 0.045 based on combined genes. Fixation index (Fst) and gene flow (Nm) indicated that there was no significant genetic differentiation occurred (Fst: 0.027, p>0.05; Nm: 17.83), which indicates a low level of genetic differentiation and a high level of gene flow between its two regional populations. The two populations did not differentiate in the neighbour-joining phylogenetic tree. In conclusion, we should strengthen the protection of species diversity.
